Abstract

¸ñÀû: ´ëÇ¥ÀûÀÎ ¸¸¼º ±¸°­³» ¿°Áõ¼º ÁúȯÀÎ Ä¡ÁÖ¿°ÀÇ Ä¡·á¿¡ Àú¿Â»ó¾Ð ÇöóÁÀÇ Àû¿ë °¡´É¼ºÀ» Æò°¡ÇØ º¸°íÀÚ, Ä¡ÁÖ Á¶Á÷³» ¸¹ÀÌ ºÐÆ÷ÇÏ°í ÀÖ´Â Ä¡Àº¼¶À¯¸ð¼¼Æ÷¿¡ ´ëÇÑ Ç׿°È¿°ú¸¦ Æò°¡Çغ¸¾Ò´Ù.

¿¬±¸ Àç·á ¹× ¹æ¹ý: µÎ¸íÀÇ È¯ÀÚÀÇ ±¸°­³»¿¡¼­ °Ç°­ÇÑ Ä¡ÀºÁ¶Á÷À» äµæÇÏ¿© primary culture ½ÃÇàÇÑ ÈÄ ½ÇÇè½Ç¿¡¼­ Ä¡Àº¼¶À¯¸ð¼¼Æ÷¸¦ °è´ë¹è¾çÇÏ¿´´Ù. ¼¼Æ÷ ½ÇÇèÀ» À§ÇØ Çï·ý °¡½º¸¦ ÀÌ¿ëÇÏ´Â Àú¿Â»ó¾Ð ÇöóÁ ÀåÄ¡¸¦ Á¦ÀÛÇÏ¿´´Ù. Ä¡ÁÖ¿°ÁõÀ» À¯µµÇϱâ À§ÇØ Porphyromonas gingivalis (Pg)ÀÇ LPS¸¦ ó¸®ÇÏ°í, CCK-8 kit¸¦ ÅëÇØ ¼¼Æ÷»ýÁ¸À² Æò°¡¸¦ ½ÃÇàÇÏ°í ¿°Áõ¼º »çÀÌÅäÄ«ÀÎÀÎ interleukin (IL)-8, IL-6ÀÇ ºÐºñÁ¤µµ¸¦ Æò°¡ÇÏ¿´´Ù.

°á°ú: Àΰ£Ä¡Àº¼¶À¯¸ð¼¼Æ÷ »ýÁ¸À² Æò°¡¿¡¼­´Â Pg LPS¸¦ ó¸®ÇÑ ±º°ú ÇöóÁ¸¦ ó¸®ÇÑ ±º, Pg LPS¿Í ÇöóÁ¸¦ ÇÔ²² ó¸®ÇÑ ±º ¸ðµÎ¿¡¼­ »ýÁ¸À²ÀÌ 92.28%¿¡¼­ 100% »çÀÌ·Î Á¸ÀçÇÏ°í Åë°èÀûÀ¸·Î À¯ÀǼºÀº °üÂûµÇÁö ¾Ê¾Ò´Ù. 6½Ã°£°ú 24½Ã°£ ¼¼Æ÷¹è¾ç½Ã°£¿¡ µû¸¥ ¼¼Æ÷»ýÁ¸À² Â÷À̵µ °üÂûµÇÁö ¾Ê¾Ò´Ù. Pg LPS¸¦ Àû¿ëÇÏ¿´À» ¶§ ´ëÁ¶±º¿¡ ºñÇØ IL-8°ú 6ÀÇ ºÐºñ°¡ Áõ°¡µÇ¾úÀ¸¸ç, Àú¿Â»ó¾Ð ÇöóÁÀÇ Àû¿ë½Ã ±× ºÐºñ°¡ Åë°èÀûÀ¸·Î À¯ÀÇÇÏ°Ô °¨¼ÒµÇ¾ú´Ù.

°á·Ð: Àú¿Â»ó¾Ð ÇöóÁ°¡ Àΰ£¼¶À¯¸ð¼¼Æ÷ÀÇ ¼¼Æ÷ »ýÁ¸À²¿¡ À¯ÀÇÇÑ ¿µÇâÀ» ³¢Ä¡Áö ¾Ê¾Ò°í, Ä¡ÁÖ¿°À¯¹ß ¿°Áõ¼º »çÀÌÅäÄ«ÀÎÀÎÀÎ IL-8°ú IL-6ÀÇ ºÐºñ¸¦ ¾ïÁ¦ÇÏ¿´´Ù.

Purpose: The purpose of this study was to evaluate the anti-inflammatory effects of non-thermal atmospheric pressure plasma (NTP) on human gingival fibroblasts (HGFs) for clinical application of periodontal treatment. Materials and
Methods: HGFs were treated with Porphyromonas gingivalis (Pg) lipopolysaccharide (LPS). Customized NTP device was developed for periodontal in vitro study. Cell viability was evaluated with cell counting kit-8. The levels of inflammatory cytokines, including interleukin (IL)-8 and 6, were determined by enzyme-linked immunosorbent assay.

Results: When NTP was applied, the cell viability did not change significantly, and there was no difference for 6 h and 24h. When Pg LPS was treated to HGFs, the secretion of IL-8 and IL-6 was increased compared to the control group. But when the NTP was applied, the secretion of them was significantly decreased.

Conclusion: NTP did not affect cell viability of HGFs. And it inhibited the LPS-induced production of IL-8 and IL-6.
